Yes, definitely!
In July, when I had to change some USD, the rate posted on the sign in most exchange offices around Nagoya Hill and Lucky Plaza was 9900 for 100USD notes. When I specified I wanted to change 500 USD in 5 notes (not a big amount for an office which manages thousands), the rate immediately became 10100, or 10150, or even 10200 in one place.
It's a big difference.
